Breckpot β-lactam synthesis

What is Breckpot β-lactam synthesis?

The Breckpot β-lactam synthesis, which involves the cyclization of esters of β-amino acids with Grignard reagent to produce β-lactam, was first reported by Breckpot in 1923.

This reaction has been adapted to create β-lactam through the use of Mukaiyama’s reagent to treat β-amino acids or N-methyl pyridinium salt to treat β-amino esters.

β-Lactams, including azetidinone, have been synthesized using this reaction.
